Chapter 1 deals mainly addresses enzymological considerations and the rational design of novel steroid-based 5aelig;-reductase inhibitors, one of which (SKF-105657)is undergoing clinical evaluation. Chapter 2 relates to the discovery, structural elucidation, anti-tumor activity and DNA cleavage properties of the calicheamicins. This continues in Chapter 3 where the authors address CC-1065 as a potent anti-tumor antibiotic with an unusual structure. Chapter 4 gives an overview on the discovery at Merck of antagonists of cholecystokinin, a peptide hormone. In Chapter 5, an elaboration of a non-classic case of finding a nonpeptide small molecule for inhibition of a peptide receptor is given. Also addressed are the structure-activity relationships for a novel series of substituted imidazoles that antagonise the angiotensin II receptor. Finally, Chapter 6 chronicles the evolution of the aldose reductase inhibitor program at Pfizer, from sorbinil to zopolrestat.
