As expected, the recovery of the receptor increased with increased rotor speed, membranes were subsequently prepared by centrifugation at 9000xg in order to obtain maximal recovery of the receptor. The stability of the crude membrane receptor was markedly improved by supplementation of the buffer with glycerol; the hormone binding activity of the membrane extract was completely retained after storage at 4#65533;C for at least 3 day. The proteolysis of the receptor (and/or nicking), by endogenous protease, had been inhibited by the inclusion of an inhibitor cocktail containing PMSF, and EDTA in extraction buffer.
